Almost everywhere I go, I always have a general medicine/first aid kit with me packed in a small wet bag. For the most part it contains: Boogie Wipes, Diaper/First aid cream, thermometer, Nasal Aspirator, Children's Tylenol and Band-Aids (not pictured)
With some extras added for camping, such as: Bug Spray, Sunscreen, Anti Itch gel (for bug bites)
Clothing:
Based on how the weather will be, depends on what to pack. For our trip, we packed for cold weather. My toddler's bag contained:
2 Pairs of Jeans, 2 pairs of Sweat Pants, 3 Long Sleeve Onesie's, 4 Long Sleeve Shirts, 3 Pairs of Fleece PJs, 3 Pairs of Socks, 2 Jackets, 1 Light Jacket, 2 Pairs of Play Shoes, 2 Hats, Diapers (or Undies) Wipes, Two Wet Bags (for dirty clothing)
Toys:
These were a must for us.
Dump Truck, Shovel & Pail, a Few Small Cars and Trucks, Bubbles and a Kid Flash Light.
We also made a scavenger hunt game to bring with us.

Stroller: Strollers can take up a lot of space in your car but if you plan to take long hikes or activities, may be good to bring along if your toddler gets tired.
Baby Carrier: Another option to bring along rather than a stroller.
Kid Play Tent: a small fold down or pop up tent for your toddler to play in.
Kid Chair: So they have their own chair to sit in.
